Allis (Alosa alosa) and Twaite (Alosa fallax) shad monitoring data sampled from the rivers Wye, Usk and Tywi using Mitochondrial DNA techniques (2013).
The river Wye, Usk and Tywi are, or form part of Special Areas of Conservation (SACs), designed under the EU Habitats Directive (92/43/EEC) for several features including the Twaite and Allis shad. The dataset contains the results from DNA analyses conducted on 226 fish eggs sampled from the river Wye, Usk and Tywi. The aim of the project was to use genetic techniques to address the weaknesses identified in the methods already adopted by Natural Resources Wales (NRW) for sampling shad eggs. The main objectives of analyses were to: - Determine whether the collected eggs belonged to Alosa spp. - Determine the proportion of Twaite and Allis shad using three different genetic markers.

Analyses of eggs collected involved DNA barcoding where a short genetic marker, usually the COI gene, of an unknown organism was sequenced using universal primers, to identify the species to which it belongs. This DNA analyses was completed in response to being unable to distinguish between the two Alosa species and reduce the risk of miss identification of fish eggs.
